Pterostilbene promotes the osteogenic differentiation of MC3T3-E1 cells inhibited by H2O2 by activating the AMPK signaling pathways

Osteoporosis is one of the common degenerative skeletal diseases whose essential mechanism imbalance between bone resorption and formation. Currently, close coordination signaling pathways at genetic level during development a hot topic osteoporosis research. The present study focused on whether pterostilbene protects against hydrogen peroxide (H2O2)-induced osteoblastic cell apoptosis, oxidative stress, reveals related underlying mechanisms. MC3T3-E1 cells were cultured treated with different concentrations H2O2 (0–1 mM) along or without pterostilbene, MTT assay Annexin V-FITC/ propidium iodide staining was applied for measuring viability apoptosis. in vitro cellular antioxidant analysis performed using 2,7-dichlorodihydrofluorescein diacetate enzyme-linked-immunosorbent serologic glutathione peroxidase malondialdehyde. In addition, alkaline phosphatase activity executed by Alizarin Red S staining. Western blot conducted to determine expression levels osteogenic-related markers, including type I collagen, osteopontin, runt-related transcription factor 2, 5′ adenosine monophosphate-activated protein kinase (AMPK) proteins. key finding that could attenuate H2O2-induced apoptosis stress. Pterostilbene also promoted osteogenic differentiation H2O2-treated through activation AMPK pathway. conclusion, blocked dysfunction, indicating its potential be promising medication treating osteoporosis.
